标题:Comparison of DNA extraction methods for muscle tissues of tuna and tuna-like species: Its implications for identification using polymerase chain reaction
摘要:This paper compares two types of DNA extraction procedures, namely, the TNES Urea and the ROSE methods for muscle tissues of tuna and tuna-like species. The tuna and tuna-like species sampled in this study were Katsuwonus pelamis, Sarda orientalis and Euthynnus affinis. An unknown fish-fillet sample that was commercially labeled as "yellow fin" was also included in the study. The presence of DNA from the samples was checked using agar rose gel electrophoresis. The result of electrophoresis after it has been stained with ethidium bromide showed that the TNES urea method had produced darker bands of DNA in the gel as compared to the ROSE method. Likewise, the DNA extracts were amplified using polymerase chain reaction (PCR) with cytochrome oxidase 1 (CO1) gene of the mitochondrial DNA as a primer. The DNA extract from fish fillet that was extracted using the TNES urea method was the only sample that showed positive band in the agar rose gel after PCR amplification. Hence, this study affirms the efficiency of the TNES urea method in extracting fish muscle tissue DNA and its subsequent usage for PCR amplification.
