  • 摘要:As label-free biomarkers, biophysical properties of cells are widely used for cell type classification. However, intrinsic biophysical markers, e.g., specific membrane capacitance (Cspecific membrane), cytoplasm conductivity (蟽conductivity) and instantaneous Young鈥檚 modulus (Einstantaneous) measured for hundreds of single cells were not yet reported. In this study, single cells in suspension (adherent cells treated with trypsin) were aspirated through a microfluidic constriction channel at 25鈥壜癈, and the entry processes and impedance profiles were recorded and translated to Cspecific membrane, 蟽conductivity and Einstantaneous. Cspecific membrane, 蟽conductivity and Einstantaneous of five cell types were quantified as 2.10卤0.38鈥壩糉鈥塩m鈭?, 0.91卤0.15鈥塖鈥塵鈭? and 5.52卤0.95鈥塳Pa for H460 cells (ncell=437); 2.52卤0.54鈥壩糉鈥塩m鈭?, 0.83卤0.12鈥塖鈥塵鈭? and 5.54卤1.04鈥塳Pa for H446 cells (ncell=410); 2.45卤0.57鈥壩糉鈥塩m鈭?, 0.99卤0.18鈥塖鈥塵鈭? and 5.16卤1.68鈥塳Pa for A549 cells (ncell=442); 1.86卤0.31鈥壩糉鈥塩m鈭?, 1.07卤0.18鈥塖鈥塵鈭? and 3.86卤0.81鈥塳Pa for 95D cells (ncell=415); 2.03卤0.35鈥壩糉鈥塩m鈭?, 0.99卤0.16鈥塖鈥塵鈭? and 3.49卤0.70鈥塳Pa for 95C cells (ncell=290). The database of Cspecific membrane, 蟽conductivity and Einstantaneous may serve as a reference for future studies of cellular biophysical properties.
