  • 摘要:Ethanol remains the leading etiological factor for the liver diseases. Some data about destructive effect of alcohol on rhythmical processes in organ-ism of mammals are known. In the present study, we aimed to evaluate the impact of chronical alco-holic intoxication within three weeks on a number of morphological and functional parameters of rats. The study of the effect of chronic alcohol in-toxication on the morphological and functional state of the liver and on a number of biochemical and hematological parameters of 6-months-old 80 male Wistar rats was made. Violation of circadian rhythms(CR) due to the action of ethanol manifests itself already at the level of diurnal dynamics of the studied micromorpho-metric parameters in the form of destruction of the CR of the hepatocyte area and NCR and changes in the characteristics of the nucleus area. We also found changes in CR for several studied biochemi-cal and hematological parameters, as well as for the rhythm of rectal temperature, the disturbance of the rhythm of which is the most obvious sign of the development of desynchronosis. It was found that steatosis develops in the liver of rats under the influence of alcohol, and changes in the level of the studied parameters are character-istic of the early stage of alcoholic liver disease. Chronic alcohol intoxication causes significant changes in the structure of biorhythms of experi-mental animals at the cellular and organ levels. The destruction of several CR, the appearance of new ones, and a significant rearrangement of the remain-ing ones and their mismatch with each other -desynchronosis -indicates a pronounced chrono-toxic effect of ethanol. CR is a daily rhythm of biological functions, with period about of 24-25 hours. Liver circadian system is controlled by a number of interacting cellular and systemic circadian mechanisms. The expression of genes that control CR is regulated by the cellular clock, rhythmic systemic,or combina-tion of both mechanisms. Alcohol demonstrates chronotoxic effect, changing or destroying CR. Destruction of CRs in liver due to chronic alcohol intake leads to changes in lipid metabolism, and to disruption in other metabolic processes in this or-gan.
