  • 摘要:Novelty statement: During Apricot in-vitro propagation a lot of difficulties were faced during the experiment (high death rate and contamination).
Therefore, this work will have the best ap-proach for the apricot establishment based on disin-fectants. In the other treatments, the survival per-centage was very low and with high contamination as compared to sodium hypochlorite 28%plus etha-nol 70% which showed a high rate of survival with a little bit of contamination. This was the first work (surface sterilization)on the selected germplasm of apricot from Gilgit Baltistan and Quetta.
The main purpose of the presented study was to investigate the techniques for in vitro surface sterili-zation of Apricot selected germplasm(Trivet, Share, Aluchi wala, Castle bright, Habi sharot, Habi gulmet, and Pana sherqila). Shoot tips of the selected plants were used as a source of explants. The shoots were previously sterilized using six treatments. The disin-fectant treatment was based on sodium hypochlorite 15,20 and 25%, Ethanol 70%,and Sodium hypo-chlorite 28% with the combination of ethanol 70%and also with double distilled water 100% and inoc-ulated in MS media supplemented with BAP 1mg/l. to eliminate microbial contamination and to increase the survival percentage of Apricot germplasm. Re-sults showed that Sodium hypochlorite 28%plus ethanol 70%(15+1min) were the best results for sur-vival percentage in Trivet(100%), Habi sharot, Gulmet,and Pana sherqila(80%),while fungal 20%, bacterial 40%, viral 40% and 60% deaths were natu-rally recorded, although in all other treatments high rate of contamination and naturally declined of ex-plants were stared after some time as compared to control.
