摘要:Psoralea corylifolia Linn., an Indian medicinal plant exhibited good antioxidant, antitumoral and immunomodulatory activities and excellent ex vivo radical scavenging capacity, which are relevant to radioprotection and uses in medicine. By the present research for the first time using EPR spectroscopy we report our investigations on “real time” levels of Ascorbate radicals (Asc.) and ROS production in organs of mice treated by Psoralea corylifolia Linn. and its biodistribution in the same organs. For the present study BALB/c mice were injected i.p. with compound (40 mg/kg), dissolved in saline. Mice were dissected at the different time intervals post injection and tissues from liver, kidneys and pancreas were homogenates in cold PBS solution and tested for organ allocation of the drug. Tissue homogenates in DMSO and DMSO solution of the spin trap N-tert-butyl-alpha-phenylnitrone (PBN) were prepared for determination of the ascorbate radicals level and for ROS products level, correspondingly. At the 4th h after treatment almost no drug concentration, was found in any of the studied organs. Maximum concentrations in livers, kidneys and pancreas were established at 90th min after drug administration. Statistical significant increased level of ROS production was found in kidneys homogenates of mice comparing to those in the livers and pancreas. The levels of ascorbate radicals in the livers and pancreas were higher than that calculated for the kidneys. In conclusion, these first EPR results characterize Psoralea corylifolia Linn. as a promising natural hepatoprotector.
