  • 摘要:PLB of Grammatophyllum scriptum (Lindl.) Bl. was used as explants. Theaim of this research was to know the influence of kinetin and NaphthaleneAcetic Acid (NAA) application on PLB differentiation and shoot growth byin vitro culture. PLB were placed on medium containing basal salt KnudsonC (KC). Research device the used is Completely Random Design (CRD) withsix treatments, every treatment with five restating. Kinetin were used incombination with NAA ((K0N0: kinetin 0 mg/l and NAA 0 mg/l (control),K1N0: kinetin 1 mg/l and NAA 0 mg/l, K2N1: kinetin 2 mg/l and NAA 1 mg/l,K3N2: kinetin 3 mg/l and NAA 2 mg/l, K4N3: kinetin 4 mg/l and 3 mg/l, K5N4:kinetin 5 mg/l and NAA 4 mg/l) to shoot induction. The data of shootinduction was examined at week 2, 4, 6, 8, and 10. The parameter examinedis amount of shoot formation, anatomy structure of PLB, shoot height,browning of shoot, and root formation. Analysis of Variance (ANOVA) wasdone to know the influence of kinetin and Naphthalene Acetic Acid (NAA)application on shoot formation and when by Duncan Multiple Range Test(DMRT) on 5% significant rate to find out the real distinction between thetreatments. The result show that the used of kinetin 1 mg/l and NAA 0 mg/lwas stimulating differentiation of PLB G. scriptum produced shoot apicaland primordial leave at third week. The used of kinetin 5 mg/l and NAA 4mg/l produced the most shoots, the highest shoot height, and rootformation.
