摘要:Attempt was made to detect the hemagglutinin (HA) genes of influenza viruses in throat swabs and washing fluid from outbreaks using the polymerase chain reaction (PCR) method. Three pairs of primers for PCR in HA genes of influenza AH1, AH3 and B viruses were prepared and examined for sensitivity and specificity. The primer of the AHI virus reacted only with genes of the AHI virus and formed a specific band (467 base pairs) by gel electrophoresis on an agarose gel with ethidium bromide staining, but not with other viruses. Also, the primers for AH3 and B viruses reacted only with genes of the AH3 and B viruses, respectively. By the present method, virus genes could be detected in 0.5PFU/5μl in cultured virus suspensions. Inhibitor (s) of PCR amplification were present in human washing fluid and reduced virus sensitivity in 1/100. Virus particles of 1-79PFU/5μl in throat swabs and washing fluid in patients made possible the detection of HA genes by PCR. PCR was shown useful for diagnosis using HA genes of influenza infection.
