摘要:The aims of this research were to know the tolerance ofAST6 lactic acid bacteria isolated from the tractus digestivus of indigenous fish to bile salt as one of probiotic condition and to know the effect of inocculvm usage from AST6 lactic acid bacteria into the resistance of the medium containing bile salt. The bile salt with the concentration of 0%,0.05%,0.10%,0.15%,and 0.20% were added to MRS medium which were done in three replicates. The growth of lactic acid bacteria were evaluated every hour based on the changes of biomass density,the decreasing of pH,and spesific growth rate. Lactic acid production were measured at the peak of exponential phase. The data obtained were analyzed with analysis of variance using Completely Randomized Design (CRD) and the different variable values affected by the treatments were analyzed by Duncan's Multiple Range Test (DMRT). Lactic acid bacteria isolated from the tractus digestivus offish were tolerance of bile salt,although there was an inhibition effect at the concentration of 0.15%. The addition of 0.15% and 0.20 % bile salt significantly (p<0.05) decreased the biomass density at the 22nd hour from 0.982 (without bile salt) to 0.802 and 0.808 respectively. The spesific growth rate reduced from 0.0974 to 0.0838,0.0765,0.05JO,and 0.0589 g cell/hour (DM bases). Lactic acid production at 21st hour significantly (p<0.05) decreased as the effect of the addition of bile salt from 0.645 to 0.477,0.361,0.376,and 0.352 mg/ml. The value of pH decreased significantly (p<0.05) up to 22 hour in all of the concentration. It could be concluded that the lactic acid bacteria isolated from the digestivus tractus of fish were tolerance of 0.15% bile salt concentration.
