  • 摘要:Dresel exhaust patucle ( <25 pum,DEP2 s) Is known to be probarbly Carcinogentc (IARC group 2A) DEP2s contauns organc compounds such as polycyeltcauomatic hydrocalbon (PAH),hetcrocyclic compounds, phenols,and ntroarenes Reactve oxygen specres (ROS) arc generated by DEP2s wthout any biologucal actvaton system Therefore,an alternative mechamusm by whuch DEP2s could be carcinogenic I8 known by the gencratuon of oxndatve DNA damage The am of thrs study was to investgate genotoxnc effects of DEP2s using sungle cell gel electrophoresis In order to evaluate the mechanusms of DEP2 genotoxtcity,the rat micro- some mednated and DNA repall enzyme treated comet assays together with routine comet assay were perfoumed DEP2s was collected from diesel engine bus dand duchlol omethane extract was obtained The organc extuact of DEP25 revealed DNA damage 1tself m NIH/3T3 cells And It showed both oxidative and mucrosome medhated DNA damages Vitamin C as an model antoxidant reduced DNA damage In endonuclasc III treated comet assay One of flavonold,galangin as a CYPIAI Inhubito1 reduced DNA damage mn the presence of S -9 mxture Our results show that DEP2 q ale gcnotoxuc and a great source of oxIdative stress,but antoxidants can signficantly reduce oxidative DNA damages And DEP2s may contan Indrecl mutagens whtch can be 1nhubrted by CYP Inhbrtors.
