摘要:Tron (Fe) 18 an esscntal metal m biologcal processes,whuch mamntans a homcostdsIs In the human body Divalent metal tuansporter 1 (DMTI) has been known as an iron transporting membrane protern,whuch 1s involved Imn the uptake Fe at the apical potion of Itestnal eputhelum,and may transport Fe across the membrane of acidified endosome In penpheral tssucs In thus study,we studed the tssue distrbution of DMTI In the Fe supplemented (FeS) duet fed rats,and the regulaton of DMTI expresson by depltng body Fe Sprague -Dawley rats were dinded mto two groups,and fed FeS (120 mg Fekg) dhet or Fe defic1ent (FeD, 2~6 mg Fe/kg) dhet for 4 wecks The evaluaton of body Fe stalus was montored by measurng sFe,UIBC and Issue Fe concentration Additonally,DMT1 mRNA levels were anulyzed in the penpheral tisues by using the quanttatve real time RT- PCR method In the FeS det fed rats,the tissue Fe was mauntauned at a relatively hugh level,and DMTI was eventually expressed In all Issues studhed DMTI was hughly expressed n the tetus,kdney and spleen,whle a moderate levels of DMTI expresson was dected In the bran,liver and heart In the digesuvc system,the hughest level ot DMT1 was found m the duodenum Feeding the FeD dhet caused a reduced body weight gan and depletion of body Fe with findng of decneased sFe,Increased UIBC and decreased tsse Fe concentration The depleton of body Fe upregulated DMT1 expression im the pernpheral tssue The expesion of DMT1 was very senstive to the body Fe depletuon In the small intestne, espectally n the duodenum. showing dramatcally hugher levels In the FeD rats than those of the FeS group In the FeD det fed anmals,the expression of DMTI was low sigmficantly 1n other tssues compared wnth the duodenum The expression of DMTI,however,was 60~ 120% hgher n the testus,kdney and spleen,and 30 一50% hugher In the lung,liver and heart,compared to the FeS duet fed rats In summary,DMT1 expression wds ubiqutous In mammaluan ussue,and the level of explesston was the organ -dependent The expression of DMTI n perpheral tsses was upregulatd by depleton of body Fe Duodenum was the most senstve tssue among orgudns studed dunng Fe depleton,and expessed the greatest level of DMTI,whle other tissucs were less higher than In duodenum Thus study supports that DMT1 plays a role 1In mantamnung the body Fe level through ntestnal uptake as well as homecostasis of Fe In the pertpheral tssue.
