摘要:Prostaglandins and endogenous cannabinoid metabolites sharethe same lipid backbone with differing polar head groups atexactly the position through which a large molecule is attachedto provide antigenicity and thus raise antisera. Hence, we hypothesizedthat antisera raised against prostaglandins linked to a largemolecule such as BSA at the carboxyl functional group wouldalso recognize endogenous cannabinoid metabolites and lead tohighly misleading interpretations of data. We found major cross-reactivityof commercial antisera raised to prostaglandins with endocannabinoidmetabolites. Furthermore, in a well-characterized cell line(WISH) or primary amnion tissue explants, endocannabinoid treatmentled to increased production of endocannabinoid metabolites asopposed to primary prostaglandins. This was apparent only afterseparation of products by thin-layer chromatography, becausethey measured as prostaglandins by radioimmunoassay.
These findings have major implications for our interpretationof data in situations in which these prostaglandin-like moleculesare formed, and they stress the need for chromatographic orspectrometric confirmation of prostaglandin production detectedby antibody-based methods.Supplementary key words ethanolamides • prostamides • endocannabinoids • anandamide
