摘要:Conjugated linoleic acid (CLA), a mixture of isomers of linoleicacid, has previously been shown to be able to decrease porcinesubcutaneous (SC) adipose tissue levels while increasing thecount of intramuscular (IM) adipose tissue in vivo. However,the underlying mechanisms through which it acts are poorly understood.The objective of this study was to investigate the differenteffects of CLA on adipogenesis in cultured SC adipose tissueand IM stromal vascular cells obtained from neonatal pigs. Asshown here, trans-10, cis-12 CLA decreased the expression ofadipocyte-specific genes as well as adipose precursor cell numbersand the accumulation of lipid in cultured SC adipose tissuestromal vascular cells. However, the cis-9, trans-11 CLA didnot alter adipogenesis in SC cultures. On the other hand, bothCLA isomers increased the expression of adipocyte-specific genesin IM cultures, together with the increasing accumulation oflipid and Oil Red O-stained cells. Collectively, these datashow that CLA decreases SC adipose tissue but increases IM adiposetissue by different regulation of adipocyte-specific gene expression.These results suggest that adipogenesis in IM adipocytes differsfrom that in SC adipocytes.Supplementary key words adipocyte-specific gene • adipose precursor cell • proliferation • differentiation • subcutaneous adipose tissue • intramuscular adipose tissue • conjugated linoleic acid
