  • 摘要:The aim of this study was to detect genetic diversity within a set of 30 wheat cultivars (Triticum aestivum L.) and another set of 30 barley genotypes (Hordeum vulgare L.) using microsatellite markers and also to establish efficiency of chosen SSR markers for the differentiation. Seventeen microsatellite markers based on PCR-reactions were used for detection of genetic diversity of the 30 wheat cultivars. These seventeen primer pairs amplified altogether 94 different polymorphic alleles with an average number of 5.5 alleles per locus. The number of alleles ranged from 3 (Xgwm95) to 9 (Xgwm174). All the seventeen microsatellite markers were polymorphic and produced reproducible data. The diversity index (DI), polymorphic information content (PIC) and probability of identity (PI) of the tested SSR markers were calculated. The diversity index of the tested SSR markers ranged from 0.347 (Xgwm495) to 0.824 (Xgwm540) with an average of 0.636. The polymorphic information content ranged from 0.332 (Xgwm495) to 0.816 (Xgwm540) with an average of 0.618. The dendrogram based on hierarchical cluster analysis using UPGMA algorithm for the detection of genetic diversity was prepared. The cultivars were grouped into two main clusters. It was not possible to distinguish cultivars Rába and Samanta from each other. Same was the case for another set of cultivars Brea and Niagara which could not be distinguished from each other.
